背景信息

MICA (MHC class I polypeptide-related sequence A) is a stress-inducible cell surface glycoprotein belonging to the MHC class I family. Unlike classical MHC class I molecules, MICA does not present antigens but functions as a ligand for the activating receptor NKG2D, expressed on natural killer (NK) cells, γδ T cells, and CD8+ αβ T cells. Its expression is typically low in healthy tissues but upregulated in response to cellular stress, infection, or malignant transformation, making it a critical mediator of innate and adaptive immune responses against infected or cancerous cells.

Structurally, MICA consists of three extracellular domains (α1. α2. α3), a transmembrane region, and a cytoplasmic tail. Polymorphisms in the MICA gene influence protein expression and receptor binding affinity, contributing to variations in immune responses. In cancer, tumor cells often evade immune surveillance by proteolytic shedding of MICA, releasing soluble forms (sMICA) that downregulate NKG2D on immune cells, thereby impairing cytotoxicity.

Recombinant MICA proteins are engineered to study these interactions and develop therapeutic strategies. Produced via expression systems (e.g., mammalian, insect cells), they retain functional epitopes for NKG2D binding and are used in immunological assays, structural studies, and as reagents to block sMICA-mediated immunosuppression. Researchers also explore recombinant MICA in immunotherapy, such as vaccines or bispecific antibodies, to enhance NKG2D-mediated tumor targeting. Additionally, recombinant MICA aids in diagnosing and monitoring diseases by quantifying sMICA levels, which correlate with cancer progression and prognosis.

Overall, recombinant MICA serves as a vital tool for understanding immune evasion mechanisms and advancing immunotherapies aimed at restoring NKG2D-dependent immune surveillance.
