The Janus kinase 3 (JAK3) is a tyrosine kinase belonging to the JAK family, primarily involved in cytokine receptor signaling. Unlike other JAKs (JAK1. JAK2. TYK2), JAK3 is predominantly expressed in hematopoietic cells and associates with the common gamma chain (γc) of receptors for interleukins such as IL-2. IL-4. IL-7. IL-9. IL-15. and IL-21. Upon cytokine binding, JAK3 activates downstream signaling pathways, including the STAT (signal transducer and activator of transcription) family, regulating immune cell development, proliferation, and function. Mutations in JAK3 can lead to severe combined immunodeficiency (SCID) due to disrupted lymphocyte signaling, underscoring its critical role in immunity.
JAK3 antibodies are essential tools for studying its expression, activation, and interactions in immune-related diseases and cancers. In research, they are used in techniques like Western blotting, immunohistochemistry, and flow cytometry to detect JAK3 levels or phosphorylation status, reflecting pathway activity. Dysregulated JAK3 signaling is implicated in autoimmune disorders (e.g., rheumatoid arthritis, psoriasis) and hematologic malignancies, making it a therapeutic target. Inhibitors like tofacitinib block JAK3 activity, validating its clinical relevance. Antibodies against JAK3 also aid in developing diagnostic assays and evaluating inhibitor efficacy, bridging basic research and therapeutic innovation.
