CD129. also known as interleukin-9 receptor (IL-9R), is a cell surface protein belonging to the type I cytokine receptor family. It consists of a ligand-specific α-chain (IL-9Rα) that pairs with the common γ-chain (γc, CD132) to form a functional receptor complex. IL-9R is primarily expressed on immune cells, including mast cells, T helper (Th2) cells, regulatory T cells (Tregs), B cells, and epithelial cells. It binds interleukin-9 (IL-9), a pleiotropic cytokine involved in allergic inflammation, immune regulation, and tumorigenesis. Signaling through IL-9R activates JAK-STAT (Janus kinase-signal transducer and activator of transcription), MAPK, and PI3K pathways, promoting cell survival, proliferation, and cytokine production.
