ACIN1 (Apoptotic Chromatin Condensation Inducer 1) is a nuclear protein involved in chromatin condensation and nuclear fragmentation during apoptosis. It was initially identified as a substrate for caspase-3. with cleavage required to activate its pro-apoptotic function. Structurally, ACIN1 contains an N-terminal nuclear localization signal, a central RNA recognition motif (RRM), and a C-terminal proline-rich domain. Beyond apoptosis, ACIN1 plays roles in RNA processing, particularly in alternative splicing, through its interaction with components of the spliceosome complex like SAP18.
Antibodies targeting ACIN1 are widely used in research to study its dual roles in cell death and RNA metabolism. These tools enable detection of full-length ACIN1 (~150 kDa) and its caspase-cleaved fragments (~65-85 kDa) in apoptosis assays. Commercial antibodies are typically validated for applications including Western blotting, immunofluorescence, and immunoprecipitation. However, users should note potential cross-reactivity with splice variants (e.g., ACIN1B) and consider post-translational modifications (e.g., phosphorylation) that may affect migration patterns in SDS-PAGE. Recent studies also explore ACIN1's involvement in neurological disorders and cancer progression, expanding its research relevance.
