Single-stranded DNA-binding protein 1 (SSB1), also known as hSSB1. is a critical component of the DNA damage response machinery in human cells. It belongs to the conserved SSB protein family, which stabilizes single-stranded DNA (ssDNA) intermediates during DNA replication, repair, and recombination. SSB1 specifically contains an oligonucleotide/oligosaccharide-binding (OB) fold domain that enables its high-affinity binding to ssDNA, protecting it from nuclease degradation or secondary structure formation.

First identified in 2010. SSB1 interacts with key DNA repair proteins like the Mre11-Rad50-Nbs1 (MRN) complex and ATM kinase, playing a role in homologous recombination repair and checkpoint activation. It localizes to sites of DNA double-strand breaks (DSBs) and facilitates the recruitment of repair factors. SSB1 also participates in maintaining genome stability by coordinating replication fork progression and resolving replication stress.

Dysregulation of SSB1 has been linked to cancer predisposition and neurodegenerative disorders.
