背景信息

**Background of Human TAS2R43 Protein**

TAS2R43. a member of the taste receptor type 2 (TAS2R) family, is a G protein-coupled receptor (GPCR) primarily expressed in taste receptor cells on the tongue. It plays a critical role in bitter taste perception, a defense mechanism to detect potentially harmful compounds. TAS2R43 is activated by specific bitter substances, such as saccharin, acesulfame-K, and certain plant-derived alkaloids. Genetic variability in the *TAS2R43* gene, particularly single-nucleotide polymorphisms (SNPs), contributes to individual differences in bitter taste sensitivity, influencing dietary preferences and aversion to toxins.

Structurally, TAS2R43 features a short extracellular N-terminus and seven transmembrane domains typical of GPCRs. Unlike other sensory receptors, TAS2Rs like TAS2R43 couple with gustducin, a G-protein that initiates signaling cascades leading to neurotransmitter release and taste signal transmission. Beyond the oral cavity, TAS2R43 is found in extra-oral tissues (e.g., gastrointestinal tract, respiratory epithelium), suggesting roles in nutrient sensing, gut motility, and innate immune responses. Its expression in airways may mediate bronchoconstriction or antibacterial responses upon bitter compound detection.

Research on TAS2R43 highlights its evolutionary significance in toxin avoidance and potential therapeutic applications, such as targeting bitter receptors to modulate metabolic or immune processes.
