背景信息

RBFOX3. also known as neuronal nuclei (NeuN), is a RNA-binding protein encoded by the *RBFOX3* gene, primarily expressed in postmitotic neurons across the central and peripheral nervous systems. It belongs to the RBFOX family of proteins, characterized by a conserved RNA recognition motif (RRM) that regulates alternative splicing and transcriptional processing of target mRNAs. RBFOX3 plays a critical role in neuronal maturation, synaptic plasticity, and maintenance of neuronal function by modulating genes involved in axon guidance, ion channel activity, and cell signaling.

The RBFOX3 antibody is a widely used tool in neuroscience research to specifically identify mature neurons. It targets the NeuN antigen, a nuclear protein epitope expressed in most neuronal cell types, excluding cerebellar Purkinje cells, retinal photoreceptors, and subsets of hypothalamic neurons. First characterized in the 1990s, this antibody has become a gold-standard marker for neuronal identification in immunohistochemistry, immunofluorescence, and Western blotting. Its applications span neurodevelopmental studies, neurodegenerative disease research (e.g., Alzheimer's, Parkinson's), and investigations into neurological disorders such as epilepsy or schizophrenia. Recent studies also explore RBFOX3's potential role in neuronal stress responses and its altered expression patterns in brain tumors, highlighting its dual utility as both a diagnostic marker and a subject of functional investigation.
