背景信息

ULBP2 (UL16-binding protein 2) is a member of the human ULBP family, which comprises MHC class I-related glycoproteins. These proteins act as ligands for the activating receptor NKG2D expressed on natural killer (NK) cells, γδ T cells, and CD8+ αβ T cells. ULBP2 is encoded by the RAET1H gene and is distinguished by its α1/α2 domain structure, enabling interaction with NKG2D to trigger cytotoxic responses against infected or transformed cells. Unlike classical MHC class I molecules, ULBP2 lacks the α3 domain and does not associate with β2-microglobulin.

ULBP2 is typically absent or weakly expressed on healthy cells but is upregulated under stress conditions, such as viral infections, DNA damage, or malignant transformation. Tumor cells often overexpress ULBP2 to evade immune surveillance, making it a biomarker of interest in cancer immunotherapy. Antibodies targeting ULBP2 are critical tools for studying its role in immune regulation and tumorigenesis. They enable the detection of ULBP2 expression in tissues, blockade of NKG2D-ULBP2 interactions to suppress pro-tumor signaling, or enhancement of immune cell activation in therapeutic strategies.

Research on ULBP2 antibodies has implications for developing cancer therapies, including antibody-drug conjugates, bispecific antibodies, or CAR-T/NK cell therapies. Dysregulated ULBP2 expression is also linked to autoimmune diseases, highlighting its dual role in immune activation and pathology. These antibodies thus serve both diagnostic and therapeutic purposes in translational immunology.
