The CASP9 (active) antibody is specifically designed to detect the activated form of caspase-9. a key protease in the intrinsic apoptosis pathway. Caspase-9 (CASP9) is an initiator caspase that exists as an inactive zymogen (pro-caspase-9) in healthy cells. Upon apoptotic stimuli, such as DNA damage or cellular stress, the intrinsic pathway is triggered, leading to mitochondrial outer membrane permeabilization and release of cytochrome c. This promotes the assembly of the apoptosome, a complex involving cytochrome c, APAF-1. and pro-caspase-9. Autocatalytic cleavage of pro-caspase-9 within the apoptosome generates active caspase-9. which subsequently activates downstream effector caspases (e.g., caspase-3/7) to execute apoptosis.
The CASP9 (active) antibody selectively recognizes the cleaved, activated form of caspase-9 (∼37 kDa fragment) or conformational changes associated with its activation, enabling researchers to distinguish it from the inactive precursor (∼46 kDa). This antibody is widely used in techniques like Western blotting, immunohistochemistry, and flow cytometry to study apoptosis mechanisms in cancer, neurodegenerative diseases, and developmental biology. Its specificity is often validated using cells treated with apoptosis inducers (e.g., staurosporine) or caspase inhibitors, as well as caspase-9-deficient models. By detecting active CASP9. researchers can assess the activation status of the intrinsic apoptosis pathway in experimental systems.
