**Background of VCP Antibodies**
Valosin-containing protein (VCP), also known as p97. is a conserved AAA+ ATPase critical for ubiquitin-dependent proteostasis. It facilitates diverse cellular processes, including endoplasmic reticulum-associated degradation (ERAD), mitochondrial quality control, DNA repair, and cell cycle regulation. VCP acts as a molecular chaperone, extracting ubiquitinated substrates from complexes or membranes for proteasomal degradation.
Antibodies targeting VCP are widely used in research to study its expression, localization, and interactions. In clinical contexts, VCP autoantibodies are rare but have been reported in some autoimmune disorders, though their pathogenic role remains unclear. More prominently, *mutations* in the *VCP* gene are linked to multisystem degenerative diseases, such as inclusion body myopathy with Paget’s disease of bone and frontotemporal dementia (IBMPFD), amyotrophic lateral sclerosis (ALS), and Charcot-Marie-Tooth disease. These mutations disrupt VCP’s ATPase activity or cofactor binding, leading to protein aggregation, organelle stress, and neurodegeneration.
VCP antibodies also serve as biomarkers in diagnosing VCP-related genetic disorders. Research continues to explore their therapeutic potential, including targeting VCP in cancers where its overexpression correlates with poor prognosis. Understanding VCP’s roles and antibody applications advances insights into proteostasis-linked diseases and treatment strategies.
