The SSX5 antibody targets the SSX5 protein, a member of the SSX (synovial sarcoma X breakpoint) family. The SSX family comprises nine paralogs (SSX1-SSX9) located on the X chromosome, initially identified through their involvement in chromosomal translocations in synovial sarcoma, where SSX1. SSX2. or SSX4 fuses with SS18. While SSX5 is less studied compared to its oncogenic counterparts, it shares structural features with other SSX proteins, including an N-terminal region with homology to Kruppel-associated box (KRAB) domains and a C-terminal divergent region rich in charged amino acids. SSX proteins are classified as cancer-testis antigens (CTAs), typically expressed in germline cells (testis, ovary) but silenced in most somatic tissues. However, SSX family members are aberrantly reactivated in various cancers, including melanoma, breast cancer, and hepatocellular carcinoma, making them potential targets for immunotherapy. SSX5-specific antibodies are primarily used in research to detect SSX5 protein expression in tumors or cell lines, aiding in understanding its role in carcinogenesis, immune evasion, or as a biomarker. Limited studies suggest SSX5 may contribute to tumor progression by modulating transcriptional regulation or interacting with chromatin modifiers.
