RCOR1 (REST Corepressor 1), also known as CoREST, is a crucial epigenetic regulatory protein that interacts with the REST (RE1-Silencing Transcription Factor) complex to mediate gene silencing. Functioning as a scaffold protein, RCOR1 facilitates the assembly of chromatin-modifying complexes, including histone deacetylases (HDAC1/2) and lysine-specific demethylase 1 (LSD1/KDM1A), collectively termed the CoREST complex. This complex targets genes involved in neuronal development, differentiation, and cellular identity by modifying histone marks to establish repressive chromatin states.

RCOR1 plays a pivotal role in neurogenesis by suppressing non-neuronal genes during stem cell differentiation into neurons.
