ROPN1 (Rhoptry Protein N1), also known as ROPN1 or cancer/testis antigen 58 (CT58), is a protein encoded by the ROPN1 gene in humans. It is primarily expressed in the testis and ciliated tissues, with roles in sperm motility and cilia function. Structurally, ROPN1 contains a conserved ROPN1 domain and an A-kinase anchoring protein (AKAP) binding region, enabling interactions with protein kinase A (PKA) regulatory subunits. This interaction is critical for organizing signaling complexes involved in regulating flagellar movement during spermatogenesis. Studies link ROPN1 to male fertility, as it stabilizes the fibrous sheath of sperm tails, ensuring structural integrity and motility. Dysregulation of ROPN1 has been associated with asthenozoospermia, a leading cause of male infertility.
Beyond reproduction, ROPN1 is identified as a cancer/testis antigen, aberrantly expressed in malignancies like lung adenocarcinoma and breast cancer, suggesting potential roles in tumor progression. Its restricted normal tissue expression makes it a candidate for cancer biomarkers or immunotherapy targets.
