背景信息

SYNE1 (Spectrin Repeat Containing Nuclear Envelope Protein 1), also known as Nesprin-1. is a giant multi-domain protein encoded by the *SYNE1* gene in humans. It belongs to the nesprin family of proteins that anchor the nucleus to the cytoskeleton by connecting the nuclear envelope to cytoplasmic actin filaments. SYNE1 is a key structural component of the linker of nucleoskeleton and cytoskeleton (LINC) complex, critical for maintaining nuclear integrity, mechanical stability, and cellular positioning during processes like cell migration and mitosis.

The protein contains spectrin repeats, a transmembrane domain, and a conserved KASH (Klarsicht/ANC-1/Syne homology) domain at its C-terminus, enabling interactions with SUN-domain proteins in the nuclear envelope. Mutations in *SYNE1* are linked to various human disorders, including Emery-Dreifuss muscular dystrophy, autosomal recessive cerebellar ataxia, and other neuromuscular pathologies, underscoring its role in tissue-specific mechanical resilience.

Recombinant human SYNE1 protein, produced via heterologous expression systems (e.g., bacteria, mammalian cells), is widely used to study its molecular interactions, nuclear-cytoskeletal coupling mechanisms, and disease-related dysfunction. Its applications span structural biology, mechanotransduction studies, and therapeutic target validation for neuromuscular and neurodegenerative conditions. Research on SYNE1 also contributes to understanding nuclear envelope dynamics in cellular aging and genome organization.
