GET4 (Guided Entry of Tail-anchored proteins 4), also known as TRC35 or C7orf55. is a critical component of the GET (Guided Entry of Tail-anchored proteins) complex, a conserved cytosolic machinery involved in the biogenesis of tail-anchored (TA) membrane proteins. TA proteins, characterized by a single transmembrane domain near their C-terminus, play essential roles in diverse cellular processes, including vesicle trafficking, apoptosis, and organelle dynamics. However, their hydrophobic C-terminal regions make them prone to aggregation in the aqueous cytosol. The GET system ensures their proper targeting and insertion into the endoplasmic reticulum (ER) membrane.

In mammals, the GET complex comprises GET3 (a targeting ATPase), GET4. GET5 (also called UBL4A), and the chaperone protein BAG6. GET4 acts as a scaffold, bridging interactions between GET3 and the BAG6/GET5 subcomplex.
