Dematin (DMTN), also known as erythrocyte membrane protein band 4.9. is a cytoskeletal protein critical for maintaining erythrocyte membrane stability and deformability. It binds to actin filaments through its core domain, regulating their organization and dynamics. Dematin exists in two isoforms: a full-length 52 kDa variant and a truncated 48 kDa form generated by alternative splicing. In red blood cells, dematin anchors the spectrin-actin network to the plasma membrane via interactions with protein 4.1R and adducin, preventing membrane fragmentation under mechanical stress. Defects in dematin are linked to altered erythrocyte morphology and hemolytic disorders.
