The DOK3 (Docking Protein 3) antibody is a research tool used to detect and study the DOK3 protein, a member of the DOK family of adaptor proteins. DOK3. also known as downstream of kinase 3. is primarily expressed in hematopoietic cells, including immune cells like B cells, T cells, and macrophages. Structurally, it contains a pleckstrin homology (PH) domain and a phosphotyrosine-binding (PTB) domain, which mediate interactions with signaling molecules such as kinases and phosphatases. Functionally, DOK3 acts as a negative regulator of signal transduction pathways, particularly those initiated by immunoreceptors (e.g., B-cell receptors, T-cell receptors) and receptor tyrosine kinases. It modulates immune responses by suppressing excessive activation of downstream effectors like MAPK/ERK and PI3K/AKT pathways, thereby maintaining immune homeostasis.
DOK3 antibodies are widely used in immunoblotting (Western blot), immunoprecipitation, and immunohistochemistry to investigate its expression, post-translational modifications, and interactions in cellular signaling contexts. Research has linked DOK3 dysregulation to autoimmune diseases, leukemias, and lymphomas, where its expression may be altered due to epigenetic silencing or mutations. For example, reduced DOK3 levels are observed in certain B-cell malignancies, suggesting a tumor-suppressive role. Unlike other DOK family members (e.g., DOK1. DOK2), DOK3 lacks specific tyrosine residues critical for recruiting inhibitory phosphatases, highlighting its unique regulatory mechanisms. Studies utilizing DOK3 antibodies continue to explore its role in immune tolerance, oncogenesis, and as a potential therapeutic target.
