背景信息

CDC2 (Phospho-Tyr15) antibodies are essential tools for studying the regulation of cell cycle progression, particularly during the G2/M transition. CDC2 (Cell Division Cycle 2), also known as CDK1 (Cyclin-Dependent Kinase 1), is a critical serine/threonine kinase that partners with cyclin B to form the Maturation-Promoting Factor (MPF), which drives mitotic entry. Its activity is tightly controlled by phosphorylation and dephosphorylation events. Phosphorylation at Tyr15. mediated by the Wee1 kinase, inhibits CDC2 activity by preventing ATP binding, thereby delaying mitotic initiation until DNA repair or other checkpoints are resolved. Dephosphorylation of Tyr15 by CDC25 phosphatases activates CDC2. enabling cell cycle progression.

Antibodies specific to CDC2 phosphorylated at Tyr15 are widely used to monitor the inhibitory phosphorylation status of CDC2 in response to DNA damage, replication stress, or checkpoint activation. These antibodies facilitate the detection of cell cycle arrest in G2 phase, making them valuable in cancer research, drug development, and studies of cell cycle regulation. Applications include Western blotting, immunofluorescence, and flow cytometry to assess checkpoint signaling pathways, evaluate chemotherapeutic agent efficacy, or explore mechanisms underlying genomic stability. Dysregulation of CDC2 phosphorylation is linked to tumorigenesis, making these antibodies relevant for biomarker discovery and therapeutic targeting in oncology.
