Phosphorylation of DREAM at Ser63 Modulates Pain Sensitivity in a Murine Neuropathic Pain Model

Chen L., et al.

Park S., et al.

Calcium-Dependent Kinase Regulation of DREAM via Ser63 Phosphorylation in Cardiac Hypertrophy

Gomez M., et al.

The DREAM (Downstream Regulatory Element Antagonist Modulator) protein, also known as calsenilin or KChIP3. is a calcium-binding protein belonging to the neuronal calcium sensor family. It functions as a transcriptional repressor by binding to downstream regulatory elements (DREs) in target gene promoters, including genes involved in pain modulation, neuronal plasticity, and apoptosis. Phosphorylation at Serine 63 (Ser63) is a critical post-translational modification regulating DREAM activity. This phosphorylation, mediated by kinases such as protein kinase A (PKA) or protein kinase C (PKC), disrupts DREAM's DNA-binding capacity, thereby relieving transcriptional repression and enabling gene expression.

The DREAM (Phospho-Ser63) antibody is a specialized tool designed to detect endogenous DREAM protein phosphorylated specifically at Ser63. It is widely used in neuroscience and molecular biology research to study calcium-dependent signaling pathways, gene regulation mechanisms, and cellular responses to stimuli like oxidative stress or neuronal injury. This antibody aids in elucidating DREAM's role in physiological processes (e.g., pain perception, memory formation) and pathological conditions, including neurodegenerative diseases, chronic pain syndromes, and cancer. Its specificity for the phosphorylated form allows researchers to distinguish activated DREAM from its unmodified state, providing insights into dynamic regulatory mechanisms. Validation typically includes Western blotting, immunohistochemistry, and immunofluorescence, ensuring reliability across experimental models.
