背景信息

The Phospho-Akt (S473) Mouse monoclonal antibody (clone 6F8) is a widely used reagent in molecular biology research to detect Akt (protein kinase B) phosphorylated at serine residue 473. a critical post-translational modification associated with its activation. Akt plays a central role in the PI3K/Akt/mTOR signaling pathway, regulating processes like cell survival, proliferation, metabolism, and apoptosis. Phosphorylation at S473. often in conjunction with Thr308. is required for full Akt activation, typically triggered by growth factors, hormones, or cellular stress.

Clone 6F8 is a mouse-derived IgG1 antibody generated using a synthetic phosphopeptide corresponding to residues surrounding S473 in human Akt1. It exhibits specificity for the phosphorylated S473 epitope across Akt isoforms (Akt1. Akt2. Akt3) in humans, mice, and rats, making it valuable for applications such as Western blotting, immunoprecipitation, and immunohistochemistry. Researchers frequently use this antibody to study oncogenic signaling, insulin resistance, or therapeutic responses in cancers and metabolic disorders, as hyperactivation of Akt is common in these conditions. Validation typically includes knockout controls or phosphatase treatment to confirm phosphorylation-dependent detection. Its reliability has established it as a staple tool for probing Akt activation status in diverse experimental models.
