The Arp2/3 complex subunit 1A (ARPC1A) is a key component of the actin-related protein 2/3 (Arp2/3) complex, a conserved macromolecular assembly critical for actin cytoskeleton remodeling. This complex facilitates the nucleation of branched actin networks, essential for cellular processes like cell motility, membrane trafficking, and cytokinesis. ARPC1A, also known as p41-ARC, is one of seven subunits forming the Arp2/3 complex, which includes ARP2. ARP3. and subunits ARPC1-5.
Structurally, ARPC1A shares homology with ARPC1B, another isoform encoded by a paralogous gene. While both isoforms participate in Arp2/3-mediated actin polymerization, they exhibit tissue-specific expression patterns, with ARPC1A broadly expressed in non-hematopoietic tissues and ARPC1B enriched in hematopoietic cells. This divergence suggests distinct regulatory roles in different cell types.
Functionally, ARPC1A stabilizes the Arp2/3 complex and enhances its interaction with nucleation-promoting factors like WASP/WAVE proteins. Dysregulation of ARPC1A has been linked to pathologies, including cancer metastasis (due to altered cell migration) and immune deficiencies. Recent studies highlight its potential role in neurological disorders and viral pathogenesis, underscoring its importance in cellular dynamics. Despite progress, mechanisms governing isoform-specific functions and post-translational modifications of ARPC1A remain active areas of research.
