ORMDL3, a member of the ORMDL (orosomucoid-like) protein family, is an endoplasmic reticulum (ER)-transmembrane protein encoded by the *ORMDL3* gene in humans. It regulates sphingolipid biosynthesis by inhibiting serine palmitoyltransferase (SPT), the rate-limiting enzyme in de novo sphingolipid production, thereby maintaining lipid homeostasis. ORMDL3 also modulates ER stress responses, calcium signaling, and the unfolded protein response (UPR), linking it to cellular stress adaptation mechanisms.  


Genome-wide association studies (GWAS) have identified *ORMDL3* polymorphisms as significant risk factors for asthma and other inflammatory diseases.
