RASAL1 (RAS Protein Activator Like 1) is a member of the RAS GTPase-activating protein (GAP) family, known for its regulatory role in the RAS signaling pathway. As a GAP, RASAL1 accelerates the hydrolysis of active GTP-bound RAS to its inactive GDP-bound form, thereby acting as a tumor suppressor by attenuating RAS-mediated cell proliferation, differentiation, and survival. Structurally, it contains conserved domains such as the N-terminal PH (Pleckstrin Homology) domain and C-terminal C2 domains, which are critical for membrane localization and calcium-dependent phospholipid binding.

Originally identified through homology searches for RAS-interacting proteins, RASAL1 has been implicated in multiple cellular processes, including cytoskeletal dynamics, calcium signaling, and apoptosis. Dysregulation of RASAL1 is linked to various pathologies. Epigenetic silencing via promoter hypermethylation has been observed in cancers like breast, ovarian, and renal cell carcinoma, contributing to RAS pathway hyperactivation and tumor progression. Paradoxically, RASAL1 may also exhibit oncogenic properties in certain contexts, such as promoting invasion in hepatocellular carcinoma.

Beyond oncology, RASAL1 plays roles in cardiovascular and fibrotic diseases by modulating fibroblast activation. Recombinant RASAL1 protein is utilized in research to elucidate its molecular interactions, post-translational modifications, and therapeutic potential.
