Recombinant human RCP (RAB coupling protein), also known as RCP9 or RAB11FIP1. is a multifunctional adaptor protein involved in intracellular membrane trafficking and signal transduction. It belongs to the RAB11-interacting protein (FIP) family, which interacts with RAB GTPases to regulate vesicular transport processes. Structurally, RCP contains an α-helical coiled-coil domain and a conserved RAB-binding domain at its C-terminus, enabling interactions with RAB11 and other RAB proteins.
Biologically, RCP plays critical roles in endosomal sorting, receptor recycling (e.g., EGFR, integrins), and maintaining cell polarity. It facilitates communication between endosomal compartments and the plasma membrane, impacting cell migration, cytokinesis, and autophagy. Dysregulation of RCP has been implicated in cancer progression, particularly in breast and ovarian cancers, where it promotes oncogenic signaling and metastasis through enhanced recycling of pro-invasive receptors.
Recombinant RCP proteins are typically produced using bacterial or mammalian expression systems, enabling in vitro studies of its molecular interactions and structural properties. Current research focuses on elucidating its post-translational modifications, tissue-specific isoforms, and potential therapeutic targeting in membrane trafficking-associated diseases.
