Regulator of G-protein signaling 22 (RGS22) is a member of the RGS protein family, which negatively regulates G-protein coupled receptor (GPCR) signaling by accelerating GTP hydrolysis on activated Gα subunits. Initially identified in 2001. RGS22 is expressed in human tissues such as the testis, prostate, placenta, colon, and pancreas. Its physiological roles remain incompletely understood, but emerging evidence links RGS22 to cancer progression. Studies show elevated RGS22 expression in colorectal cancer, hepatocellular carcinoma (HCC), and pancreatic ductal adenocarcinoma, where it appears to function as a context-dependent tumor modulator. In colorectal cancer, RGS22 may suppress Wnt/β-catenin signaling and promote apoptosis, while in HCC, it has been associated with enhanced tumor invasion and metastasis through unclear mechanisms.
