Caspase-1 (CASP1), also known as interleukin-1β converting enzyme (ICE), is a cysteine-aspartic protease that plays a central role in inflammasome-mediated inflammatory responses and pyroptosis. It is synthesized as an inactive zymogen (pro-CASP1) and undergoes proteolytic activation upon sensing pathogenic or stress signals, typically through inflammasome complexes like NLRP3. Activated CASP1 cleaves pro-inflammatory cytokines pro-IL-1β and pro-IL-18 into their mature forms, which drive systemic inflammation.
