Interferon alpha-inducible protein 6 (IFI6), also known as G1P3. is a small, highly conserved interferon-stimulated gene (ISG) product that plays a critical role in cellular responses to viral infections and stress conditions. It was first identified in the 1990s as part of the interferon (IFN)-α/β signaling pathway, which is central to innate immunity. IFI6 is encoded by a gene located on human chromosome 1p35 and consists of two exons, producing a 6.9 kDa protein with a hydrophobic N-terminal region and a conserved C-terminal domain.
Functionally, IFI6 exhibits antiviral properties, particularly against RNA viruses like hepatitis C virus (HCV) and human immunodeficiency virus (HIV). It localizes to the endoplasmic reticulum (ER) and mitochondria, where it modulates ER stress responses and inhibits apoptosis by stabilizing mitochondrial membranes. This antiapoptotic activity enables infected cells to survive longer, paradoxically facilitating viral persistence in some contexts. For example, IFI6 is hijacked by HCV to promote chronic infection by delaying host cell death.
Recombinant IFI6 protein is produced using expression systems (e.g., E. coli or mammalian cells) for structural and functional studies. Its applications span virology research, drug discovery for viral diseases, and investigations into cancer biology, as IFI6 overexpression has been observed in breast and ovarian cancers. Recent studies also explore its role in COVID-19 pathogenesis and autoimmune disorders. Despite progress, the precise molecular mechanisms underlying IFI6's dual roles in antiviral defense and pathology remain an active area of research, highlighting its complexity in cellular homeostasis and disease progression.
