背景信息

CD14 is a glycosylphosphatidylinositol (GPI)-anchored cell surface protein predominantly expressed on monocytes, macrophages, and neutrophils. It functions as a co-receptor for bacterial lipopolysaccharide (LPS) by binding to LPS-LBP (lipopolysaccharide-binding protein) complexes and transferring them to the TLR4/MD2 receptor complex, initiating innate immune responses. CD14 exists in membrane-bound (mCD14) and soluble (sCD14) forms, both critical in modulating inflammatory signaling.

CD14 antibodies are immunological tools targeting specific epitopes of the CD14 protein. Monoclonal antibodies (e.g., clones 61D3. MEM-18) are widely used in research and diagnostics to identify monocytes/macrophages via flow cytometry, immunohistochemistry, or Western blot. These antibodies help study CD14's role in LPS signaling, phagocytosis, and immune cell differentiation. Additionally, anti-CD14 antibodies have therapeutic potential; blocking CD14 can attenuate LPS-induced inflammation, making them candidates for treating sepsis or chronic inflammatory diseases.

However, CD14's dual role in pro- and anti-inflammatory pathways complicates therapeutic targeting. Soluble CD14 also serves as a biomarker in conditions like sepsis, rheumatoid arthritis, and cardiovascular diseases. Recent research explores CD14's interaction with non-bacterial ligands (e.g., DAMPs) and its influence on adaptive immunity. Despite challenges, CD14 antibodies remain vital in elucidating innate immunity mechanisms and developing precision therapies.
