Endothelin-3 (EDN3) is a 21-amino acid peptide belonging to the endothelin family, which includes EDN1. EDN2. and EDN3. These peptides bind to G protein-coupled receptors (EDNRA and EDNRB) to regulate diverse physiological processes, including vasoconstriction, cell proliferation, and neural crest cell development. EDN3 plays a critical role in the enteric nervous system (ENS) formation, melanocyte development, and cardiovascular function. Dysregulation of EDN3 signaling is linked to congenital disorders like Hirschsprung’s disease (characterized by aganglionic megacolon) and Waardenburg syndrome, as well as cancers such as melanoma and neuroendocrine tumors.
