CARD11 (Caspase Recruitment Domain Family Member 11) is a scaffolding protein critical for antigen receptor-mediated signaling in lymphocytes. It belongs to the membrane-associated guanylate kinase (MAGUK) family and contains a caspase recruitment domain (CARD), a coiled-coil domain, and a PDZ domain. CARD11 plays a pivotal role in activating the NF-κB signaling pathway, which regulates immune cell proliferation, survival, and cytokine production. Upon antigen receptor engagement, CARD11 forms a multiprotein complex with BCL10 and MALT1 (the CBM complex), facilitating downstream NF-κB activation.
Dysregulation of CARD11 is linked to immune disorders and cancers. Germline gain-of-function mutations cause B-cell lymphoproliferative disorders or severe immunodeficiency, while loss-of-function mutations result in primary immunodeficiencies like severe combined immunodeficiency (SCID). Somatic mutations in CARD11 are associated with diffuse large B-cell lymphoma (DLBCL) and other lymphoid malignancies, often leading to constitutive NF-κB activation and uncontrolled cell growth.
CARD11 antibodies are primarily used in research and diagnostics to detect protein expression or mutation status in immune cells. In clinical contexts, they aid in identifying genetic aberrations in immunodeficiencies or lymphomas. Therapeutic targeting of CARD11-related pathways, including inhibitors of the CBM complex or NF-κB, is under investigation for treating autoimmune diseases and cancers. However, direct therapeutic antibodies against CARD11 remain experimental, with current focus on small-molecule inhibitors modulating its signaling cascade.
