Caspase-12 (CASP12) is a member of the cysteine-aspartic protease (caspase) family, primarily implicated in apoptotic pathways and inflammatory responses. Unlike other caspases, human CASP12 exists in an inactive truncated form in most populations due to a genetic polymorphism (rs497116) that introduces a premature stop codon. This inactivation is thought to confer evolutionary advantages by reducing susceptibility to excessive inflammatory reactions, such as sepsis. However, some individuals of African descent retain the ancestral full-length, functional CASP12. which may increase vulnerability to inflammatory disorders.
CASP12 antibodies are tools used to detect and study the expression, localization, and function of this protease. These antibodies are critical in research exploring its role in endoplasmic reticulum (ER) stress-induced apoptosis, regulation of pro-inflammatory cytokines (e.g., IL-1β), and associations with neurodegenerative diseases (e.g., Alzheimer’s) or autoimmune conditions. Species-specific antibodies are essential, as murine CASP12 (functional in mice) differs structurally from the human variant. Applications include Western blotting, immunohistochemistry, and flow cytometry to analyze tissue or cell-specific expression. Researchers must verify antibody specificity due to homology with other caspases and the prevalence of CASP12 pseudogene transcripts. Understanding CASP12's dual roles in cell death and inflammation continues to drive interest in therapeutic targeting, making these antibodies valuable for mechanistic and diagnostic studies.
