背景信息

The Kir6.2 (Phospho-Thr224) antibody is a specialized tool used to detect the phosphorylated form of the Kir6.2 potassium channel subunit at threonine 224 (Thr224). Kir6.2. encoded by the KCNJ11 gene, is a critical component of ATP-sensitive potassium (KATP) channels, which link cellular metabolic status to membrane excitability. These channels are hetero-octameric complexes composed of four pore-forming Kir6.2 subunits and four regulatory sulfonylurea receptor (SUR) subunits. Phosphorylation at Thr224. located in the cytoplasmic C-terminal domain of Kir6.2. is a key post-translational modification that modulates channel activity. This site is targeted by protein kinases such as protein kinase A (PKA) and protein kinase C (PKC), influencing channel gating and sensitivity to ATP. Phosphorylation at Thr224 generally reduces channel open probability, promoting membrane depolarization and insulin secretion in pancreatic β-cells. The Kir6.2 (Phospho-Thr224) antibody is widely used in research to study KATP channel regulation in metabolic and cardiovascular contexts, particularly in diabetes, hyperinsulinism, and cardiac ischemia. It enables detection of phosphorylation-dependent signaling pathways affecting insulin release, vascular tone, and myocardial protection. Validation typically includes Western blotting, immunohistochemistry, and immunofluorescence in tissues/cells expressing Kir6.2. such as pancreatic islets, cardiac muscle, or brain. Dysregulation of Kir6.2 phosphorylation is implicated in diseases like neonatal diabetes and DEND syndrome, making this antibody valuable for mechanistic and therapeutic investigations.
