CD158D, also known as KIR2DL4 (killer cell immunoglobulin-like receptor 2DL4), is a member of the immunoglobulin-like receptor (KIR) family expressed primarily on natural killer (NK) cells and a subset of T lymphocytes. Discovered in the 1990s, KIR receptors regulate NK cell activity by interacting with human leukocyte antigen (HLA) molecules. Unlike most inhibitory or activating KIRs, KIR2DL4 exhibits dual functionality due to its unique structural features: a single extracellular immunoglobulin domain, a transmembrane region, and a cytoplasmic tail containing both inhibitory (ITIM) and activating motifs.
