ARHGEF5 (Adenylate Rho Guanine Nucleotide Exchange Factor 5), also known as TIM1 or ephexin-1. is a member of the Dbl family of Rho guanine nucleotide exchange factors (GEFs). These proteins activate Rho GTPases by catalyzing the exchange of GDP for GTP, enabling their participation in diverse cellular processes. Structurally, ARHGEF5 contains conserved DH (Dbl homology) and PH (Pleckstrin homology) domains critical for its GEF activity, as well as a transmembrane region that facilitates localization to the plasma membrane.
