背景信息

ARRDC3 (Arrestin Domain-Containing Protein 3) is a member of the α-arrestin family, a subgroup of arrestin-related proteins involved in regulating endocytic trafficking, signaling, and ubiquitination-mediated degradation of membrane receptors. Unlike β-arrestins, which primarily modulate G protein-coupled receptor (GPCR) desensitization, α-arrestins like ARRDC3 interact with E3 ubiquitin ligases (e.g., NEDD4. WWP2) through conserved PPxY motifs, facilitating substrate ubiquitination and lysosomal or proteasomal degradation. ARRDC3 contains two N-terminal arrestin domains and C-terminal motifs critical for mediating protein-protein interactions.

Functionally, ARRDC3 regulates diverse cellular processes, including metabolic homeostasis, cell migration, and tumor suppression. It modulates insulin signaling and adipocyte metabolism via its role in degrading β2-adrenergic receptors (β2-ARs) and integrins, impacting obesity and diabetes pathways. In cancer, ARRDC3 acts as a tumor suppressor by promoting oncoprotein degradation (e.g., NOTCH1) or inhibiting metastasis-related pathways.
