RSAD1 (Radical S-Adenosyl Methionine Domain-Containing Protein 1), also known as viperin or CIG5. is an interferon-inducible protein that plays a critical role in innate immunity against viral infections. First identified for its antiviral activity, RSAD1 is conserved across vertebrates and expressed in various tissues, particularly in immune cells. It functions by disrupting viral replication cycles through multiple mechanisms, including enzymatic activity linked to its radical S-adenosylmethionine (SAM) domain. This domain enables RSAD1 to catalyze radical-mediated reactions, producing antiviral molecules like ddhCTP, which inhibits viral RNA polymerases.

Structurally, RSAD1 contains an N-terminal amphipathic helix for membrane association, a central radical SAM domain, and a variable C-terminal region. Its expression is strongly upregulated by type I and III interferons, cytokines released during viral infections.
